脱管散对创伤修复影响的实验研究 被引量:8

摘要 目的:以脱管散为载体,通过观察脱管散对大鼠创伤修复模型创面肉芽组织中相关生长因子及细胞外基质表达的影响,试图从分子水平探讨中药外用制剂促进创伤修复的机制。方法:采用经典创伤修复模型,收集创面肉芽组织中不同时期的肉芽组织,经过组织芯片技术处理后,用SABC法检测创面中bFGF、Fn的表达,高倍镜下病理切片观察,采用BI-2000图像分析系统进行定量灰度分析。结果:大鼠创面肉芽组织生长旺盛,创面肉芽组织中细胞所表达的生长因子分布存在差异,在造模后6d治疗组(脱管散组)bFGF表达与阳性对照组、空白对照组存在组间显著性差异(P<0.01);造模后6、14d治疗组Fn表达与阳性对照组、空白对照组存在组间差异(P<0.05)。结论:脱管散能够通过刺激创面内源性bFGF、Fn增加,进而促进创伤的修复。 Objective: To study the mechanism of external application T. C. M of accelerating wound repair by observing the expression of growth factor and extracellular matrix in the granulation tissue on the basic of rat wound repair model treated by TuoGuanSan. Methods: The classical wound repair model was Full-thickness excisional wound on the back of the rats and collected the granulation tissue of the wound fields at post wounding days 6,10,14. Paraffin'section was made as the tissue chip. The basic Fibroblast growth factor (bFGF)and Fibronectin(Fn) expression in granulation tissue were observed by StreptAvidin-Biotin-Complex Immunohistochemistry in various group, and the picture by BI-2000 system was analyzed at end. Results: The results showed that the expression of the bFGF and Fn in wound fields were different in various groups. There were a significant differene of the bFGF(P〈0.01) at post wounding 6 days and the Fn(P〈0.05) at post wounding 6 or 14 days among various groups. Conclusion: TuoGuan,San can stimulate the cells of wound fields to produce some growth factors from itself strengthen the function of cells,which may be the mechanism of T. C. M accelerate wound repair.
