芪众颗粒预防流行性感冒的实验研究

Experimental observation of Qi-zhong granules in preventing influenza

目的了解芪众颗粒对大鼠含药血清中IL-2、IFN-r水平的干预和对甲型流感病毒A3的抑制作用,探讨芪众颗粒的作用机制。方法36只大鼠随机分为6组:芪众颗粒组低、中、高剂量组,玉屏风颗粒组、病毒唑组及空白对照组。制备各组大鼠含药血清,用双抗体夹心ABC-ELISA法检测各标本血清中的IL-2、IFN-r水平;采用预防给药的方式和观察犬肾细胞即MDCK细胞病理变化(cytopathic effect,CPE)的方法,计算流感细胞所致CPE的抑制率。结果与空白对照组比较,各药物组IL-2和IFN-r均有提高(P<0.05或P<0.01);其中IL-2以芪众低剂量组、玉屏风组和病毒唑组较高,三者之间比较,P>0.05;IFN-r以芪众高剂量组最高,与玉屏风组、病毒唑组比较,P均<0.05。抗流感病毒实验中各组均无明显抑制病毒的作用。结论芪众颗粒能提高大鼠IL-2和IFN-r水平,从而增强机体的免疫功能;芪众颗粒无明显的抗甲型流感病毒A3的作用;提示芪众颗粒有可能通过调节机体免疫力或干扰病毒免疫病理等途径达到预防流感的目的。

Objective To evaluate the effect of Qi-zhong granules on the intervention of IL-2 and IFN-r level in rat sera with the drug and its action on influenza virus A , and to investigate its mechanism of Qi-zhong granules. Methods Thirty six rats were randomly derided into six groups： three groups treated with Qi-zhong granules respectively with low, middle and high doses, a controlgroup with herbal medicine Yu-ping feng granules, a control group with virazole and a blank group. Rat sera with the drug were detected IL-2 and IFN-r levels by double antibody ABC-ELISA. The method of preventive administration was adopted. The cytopathic effect of MIDCK cells（CPE） was observed to calculate the inhibition ratio of CPE caused by influenza cells. Results Compared with the blank group, both IL-2 and IFN-r levels were boosted in drug treated groups （P 〈0.05 or P 〈 0.01 ）. IL-2 levels were higher in groups with low lose of Qi-zhong granula, Yu-ping feng granules treated group and virazole treated group. There was no significant differences above three groups （P 〉0.05）. The highest level of IFN-r appeared in the high lose of Qi-zhong granules treated group, There are significant differences as compared with the group treated by Yu-ping feng granules and virazole （P 〈 0.05）respectively.In a test of anti influenza virus, the administration groups were of no remarkable effect on inhibiting virus. Conclusion Qi-zhong granules can boost levels of IL-2 and IFN-r in rats, thus enhancing body＇ s immune function. However, Qi-zhong granules have no significant effect in inhibiting influenza A virus, It is indicated that Qi-zhong granules may have effect on prevention of influenza by regulating body＇s immunity or interfering immunopathogenesis of virus.
