摘要
目的研究豪猪蛋白(Hedgehog)信号途径中Smo蛋白在人结肠癌组织中的表达,初步探讨Smo与结肠癌血管生成的关系。方法用免疫组化方法研究人结肠癌组织中Smo蛋白的表达;同时用CD34单克隆抗体标记结肠癌组织切片中新生血管,计算微血管密度(micro-vascular density,MVD),并统计Smo蛋白的表达与MVD的关系。结果在总共39例结肠癌标本中,Smo阳性24例,阴性15例,阳性率61.54%;MVD平均值68.70±11.65,Smo阳性组MVD均值71.25±12.56,Smo阴性组MVD均值64.05±8.88,两组统计分析差别有统计学意义(t检验,P<0.05)。结论Smo蛋白在人结肠癌中活化,并参与了结肠癌的发生发展。促进新生血管的生成是其可能的机制之一。
Objective To study the expression of smoothened protein, a component of hedgehog signal pathway, and its relationship with angiogenesis in human colon cancer. Methods The expressions of smoothened protein were investigated by immunohistochemical method in 39 cases of human colon cancer tissues and microvascular density (MVD) were examined by using immunohistochemical CD34 staining. Results The positive expression rate of smoothened protein was 61.54% (24/39) in 39 cases. The average MVD of total 39 cases was 68.70 ± 11.65 and the MVD of positive group (71.25 ± 12.56) increased significantly than that of negative group (64.05 ± 8.88) (P〈0.05). Conclusion Smoothened protein is activated and may play an important role in development of colon cancer. Promoting angiogenesis might be one of its mechanisms.
