摘要
目的探讨甘利欣对体外诱导的酒精性脂肪肝影响。方法以人正常肝细胞株L02为实验材料,采用乙醇诱导建立酒精性脂肪肝的离体细胞模型,后用甘利欣处理。流式细胞仪检测作用前后细胞周期的改变;分别用ALT、AST、GGT试剂盒测定谷丙转氨酶(ALT)、谷草转氨酶(AST)以及γ-谷氨酰转肽酶(GGT)的泄露量。结果25μg/ml甘利欣可使酒精性脂肪肝细胞的S期细胞明显减少(P〈0.05),并且使ALT、AST、GGT的泄露量明显降低(P〈0.05)。结论实验结果证明,甘利欣能减缓酒精性脂肪肝细胞的增殖生长速度,并能降低酒精性脂肪肝细胞ALT、AST及GGT的泄露量,从而减轻酒精性脂肪肝的肝功能损害。
Objective To study whether Diammonium glycyrrhizinate iniection(DGI)has therapeutical effect On induced alcohol-ic fatty liver in vitro. Methods Flow cytometry assay was used to detect the diversity of cell cycle, and follow the introduction in the kit to measure. the releasing quantity of alanine aminotransferase(ALT). aspartate aminotransferase(AST)and 7-glutamyhrans-ferase(GGT).Results After treated with DGI at a concentration of 25μg / ml, the steatosis liver cells at S phase decreased remarkably(P〈0.06), furthermore the releasing quantity of ALT, AST and GGT degraded significantly(P〈0.05).Conclusion DGI could not only make the proliferation speed of alcoholic fatty liver ceils postponed, but also reduce the releasing quantity of ALT, AST and GGT, accordingly relieve the liver function damage.
