冠心II号系列组方对犬急性心肌缺血保护作用的比较研究

Comparative study on the protective effects of different Guanxin II Formula(冠心II号方) on acute myocardial ischemia in dogs

目的:比较冠心II号不同组方对犬急性心肌缺血的保护作用。方法:采用结扎犬冠状动脉前降支的方法造成犬急性实验性心肌缺血模型。描记心外膜电图,计算心肌缺血程度。硝基四氮唑蓝(N-BT)组织化学染色方法测定心肌梗死面积。放射免疫法测定血浆内皮素(ET)和6-酮前列腺素F1α(6-Keto-PGF1α)、血栓烷素(TXB2)活性的变化。同时测定冠脉血流量。结果:冠心II号系列组方均具有显著改善犬急性心肌缺血和心肌梗死的作用。在减轻心肌缺血程度方面,各组作用强度基本相当。但微米制剂及汤剂给药组在减小心肌梗死面积、增加冠脉血流量、抑制血浆ET活性、增加血浆6-Keto-PGF1α/TXB2比值方面,较有效组分重组复方、血分药有效组分重组复方、气分药有效组分重组复方作用强,且在作用持续到180分钟时部分作用与血分药、气分药有效组分重组复方组有显著性差异。微米制剂与汤剂作用强度相当。有效组分重组复方疗效优于气分药、血分药重组复方,但无明显差异。结论:冠心II号系列组方均有明显对抗犬心肌缺血的作用,但各组药物作用强度有一定差别。

Objective：To compare the protective effects of different GUANXIN Ⅱ formula on acute myocardial ischemia in dogs. Methods： The acute ischemia models were set up by ligation of left ventricular anterior artery （LAD）. The epicardial electrogram was used to measure the myocardial ischemia degree. Quantitative histological assay （ nitroblue tetrazolium, N - BT stain） was used to determine the area of myocardial infarction. And radio immunological assay was used to measure the changes of the ET and the ratio of 6 - Keto - PGF1α/ TXB2. Meanwhile, the coronary blood flow was determined. Results： The different GUANXIN Ⅱ formula could obviously alleviate damages caused by myocardial ischemia and had similar effect in relieving myocardial ischemia degree. However, micrometer praeparatum and medicinal broth group had a stronger effect in reducing the ischemic area, inhibiting the ET activity and increasing the ratio of 6 - Keto - PGF1α/ TXB2 than the other groups. Moreover, when the effect continued to 180 min, there were statistic difference between the micrometer praeparatum and medicinal broth group and the Qifen（气分）, Xuefen（血分） groups. Meanwhile, micrometer praeparatum and medicinal broth group had a similar effect ; active principle recombination group had a better effect than Qifen （气分）, Xuefen （血分） groups, but had no significant difference. Conclusion： Different GUANXIN Ⅱ formula had obvious inhibition effect on the damages caused by myocardial ischemia, but their effects were in different degree.
