金银花总黄酮对小鼠免疫性肝损伤的保护作用 被引量:18

摘要 目的:观察金银花总黄酮(Lonicera gaponica total flavone,LJTF)对卡介苗(BCG)联合脂多糖(LPS)所致小鼠免疫性肝损伤的保护作用。方法:采用BCG 2.5mg静脉注射致敏小鼠,d10给于LPS 7.5μg静脉攻击造成小鼠免疫性肝损伤模型,ig给于LJTF(100、200、400mg/kg)连续10天。分光光度法测定血清ALT、AST浓度,肝匀浆中MDA、SOD的含量;免疫组化研究肝脏肿瘤坏死因子-α(TNF-α)和核因子(NF)-κB p65的表达。结果:免疫性肝损伤小鼠血清ALT、AST较正常组明显升高,肝脏中MDA明显升高而SOD较正常组明显降低。LJTF ig能降低免疫性肝损伤小鼠血清中升高的ALT、AST含量,降低肝匀浆中MDA水平,同时升高其低下的SOD水平,LJTF也能降低免疫性肝损伤小鼠TNF-α和NF-κB p65在肝脏中的强烈表达。结论:LJTF对免疫性肝损伤小鼠有保护作用,机制与减少自由基的产生、抑制细胞膜脂质过氧化、减少炎症介质的释放有关。 Objective:To study the protective effect of LJTF on immunological liver injury in mice. Methods:The model of mouse immunological liver injury was induced by injection of Bacillus Calmette Guefin (BCG) and lipopolysaccharide (LPS) from tail vein. The alanine amin- otransferase (ALT) ,aspartate aminotransferase (AST) level in serum and alondiadehyde (MDA) content, superoxide dismutase (SOD) activities in liver were assayed by spectrophotometry. TNF - α and NF - κB p65 in hepatic tissues was also determined by the method of immunohistochemistry to evaluate the protective effect of LJTF. Results: Immunological liver injury was successfully induced by BCG + LPS. LJTF ( 100,200,400 mg/kg) ig x 10d significantly decreased ALT,AST activity in serum, reduced liver MDA content in immunological liver injury mice. LJTF also improved the reduced SOD level in liver homogenate. Further more, LJTF could inhibit the express of TNF - α and NF - κB p65 in liver tissue. Conclusion : LJTF showed significantly protective action on immunological liver injury in mice. Its mechanism possibly due to deplete ORF, suppressor cell lipid peroxidation and diminish mediators of inflammation.
