摘要
目的:观察加减地黄饮子对β-淀粉样蛋白1-40(Aβ1-40)诱导的AD模型大鼠大脑海马组织AchE和NOS1表达的影响。方法:采用大鼠海马立体定向注射凝聚态Aβ1-40诱导老年性痴呆(Alzheimer's disease,AD)动物模型。采用免疫组织化学染色法和计算机图像分析技术测定海马区AchE和NOS1表达。结果:模型组(6.42±0.38)AchE表达与假手术组(6.74±0.47)比较无显著性差异。与模型组比较,加减地黄饮子组(3.77±0.29)AchE表达明显减少,作用效应较盐酸多奈哌齐(2.25±0.20)弱。与假手术组(5.74±0.39)相比,模型组大鼠海马区NOS1表达下降(2.38±030),与模型组比较,地黄饮子组NOS1的表达增加(4.43±0.40),作用优于盐酸多奈哌齐(3.56±0.27)。结论:加减地黄饮子可通过抑制AD模型大鼠海马组织AchE和上调AD模型大鼠海马组织NOS1表达而发挥抗AD的作用。
Objective: To observe the effects of Modified Decoction of Rehmanniae on expression of Ache 和 NOS1 protein in hippocampal in Alzheimer's disease rats induced by amyloid - β1 - 40 injection into brain. Methods: AD model in rats was induced by amyloid - β1 - 40 sterotaxis injection the bilateral hippocampus. Modified Decoction of Rehmanniae was geven by oral administration for 28 days. the expression levels of AchE 和 NOS1 protein in the hippocampus were measured by immunohistochemical SABC techniques. Results: the levels of AchE expression from t the Modified Decoction of Rehmanniae group ( 3.77 ± 0.29) was significantly lower than that in the model group (6.42 ± 0.38, p 〈 0.05 ) , and the levels of NOS1 expression from the model group (2.38 ± 030) was significantly Lower than that in the sham group ( 5.74 ± 0.39 ) , and that from the Modified Decoction of Rehmanniae group (4.43 ± 0.40 ) was significantly highter than in the model group ( p 〈 0.05 ). Conclusions: Modified Decoction of Rehmanniae has sighificant inhibitory effects on expression of AchE and Up - regulateing NOS1 in hippocampus of AD rats, and could be used as an effective drug for AD
