p27kip1基因转染联合化疗对胃癌细胞生长的影响

摘要 背景:p27kipl是一种抑癌基因,早期研究显示p27kipl基因转染能明显抑制胃癌、食管癌细胞生长.表明该基因疗法可能是治疗胃癌、食管癌的新途径,深入阐明p27kipl的抑癌机制,可为p27kipl基因治疗胃癌、食管癌提供可靠的理论基础。目的:研究p27kipl基因转染联合化疗对胃癌细胞生长的影响。方法:将p27kipl重组腺病毒(Ad-p27kipl)转染胃癌细胞SGC-7901,免疫细胞化学法检测p27kipl的表达。加入5-氟尿嘧啶(5-FU)和顺铂(DDP)后.甲基噻唑基四唑(MrIT)法测定细胞生长抑制率,^3H-胸腺嘧啶核苷(^3H-TdR)掺入试验测定DNA的合成。结果:Ad-p27kipl转染胃癌细胞后,p27kipl表达明显增强。加入5-FU和DDP后,细胞生长抑制率显著增高.^3H-TdR掺人量显著降低(P〈0.01)。结论:p27kipl基因转染联合化疗能明显抑制胃癌细胞的生长。 Background:p27kipl is an anti-oncogene. Early studies have demonstrated that p27kipl gene transfer has significant inhibitory effect on growth of esophageal and gastric carcinoma cells. These results indicate that this gene transfer may be a novel strategy for the treatment of esophageal and gastric carcinoma, and clarification of its mechanism may provide theoretical basis for further p27kipl gene therapy. Aims: To investigate the effect of p27kipl gene transfer combined with chemotherapy on growth of gastric carcinoma cells. Methods: Recombinant adenovirus carrying p27kipl gene (Ad-p27kipl) was transfected into gastric carcinoma cells SGC-7901. Expression of p27kipl was detected by immunocytochemical technique. After treated with 5-fluorouracil (5-FU) and cisplatin (DDP), respectively, growth inhibitory rate of gastric carcinoma cells was evaluated by methyl thiazolyl tetrazolium (MTT) assay, DNA synthesis of cells was determined by 3H-thymidine (3H-TdR) incorporation. Results: After transfected with Ad-p27kipl, expression of p27kipl of gastric carcinoma cells increased significantly. Growth inhibitory rate of gastric carcinoma cells treated with 5-FU and DDP increased obviously and 3H-TdR incorporation decreased (P〈0.01). Conclusions: p27kipl gene transfer combined with chemotherapy can obviously inhibit the growth of gastric carcinoma cells.
