背景:BRCAA1蛋白是一个新蛋白,其在胃癌发生、发展中的作用尚不明确。目的:探讨BRCAA1蛋白在胃癌组织与不同分化细胞株中的表达水平及其临床意义。方法:收集60例胃腺癌患者,分别取胃癌和癌旁组织,应用免疫组化SP法检测BRCAA1蛋白的表达水平,并分析其与胃癌分级的关系。蛋白质印迹分析检测BRCAA1蛋白在胃癌高分化(MKN-1和MKN-74)、中分化(SGC-7901)和低分化(KATO-Ⅲ和MGC80-3)细胞株中的表达水平。结果:胃癌组织中BRCAA1蛋白的表达阳性率(23/60,38.3%)显著高于癌旁组织(0/60,0%)(P<0.01);BRCAA1蛋白在高分化(2/17,11.8%)、中分化(11/26,42.3%)和低分化胃癌组织中(10/17,58.8%)的表达阳性率存在显著差异(P<0.05)。蛋白质印迹分析结果表明BRCAA1蛋白在胃癌高、中、低分化细胞株中的表达水平逐渐增强。结论:BRCAA1蛋白参与了胃癌的发生、发展,其表达水平与胃癌低分化程度密切相关,BRCAA1蛋白可能是一个胃癌不良预后的标志物。
Background:BRCAA1 is a novel protein and its role in development of gastric cancer is not yet clear. Aims: To investigate expression and its clinical significance of BRCAA1 protein in gastric cancer tissue and various differentiated cell lines. Methods: Sixty patients with gastric adenocarcinoma were involved, cancerous and paracancerous tissues were taken. The expression of BRCAA1 was detected by immunohistochemical SP staining method. The correlation between expression of BRCAA1 and pathological grading of gastric cancers was analyzed. Western blot was used to analyze the expression of BRCAA1 protein in well differentiated (MKN-1 and MKN-74), moderately differentiated (SGC-7901) and poorly differentiated (KATO-m and MGCS0-3) cell lines. Results: The positivity rate of BRCAA1 protein expression in gastric cancerous tissue (23/60, 38.3%) was significantly higher than that in paracancerous tissue (0/60, 0%) (P〈0.01). There were significant differences of BRCAA1 protein expression in well differentiated (2/17, 11.8%), moderately differentiated (11/26, 42.3%) and poorly differentiated gastric cancer tissues (10/17, 58.8%) (P〈0.05). Western blot analysis showed that expression of BRCAA1 protein was gradually increased with the cell line differentiation degrees decreased. Conclusions: BRCAA1 protein is involved in the development of gastric cancer, and its expression is closely associated with poorly differentiated gastric cancer. BRCAA1 may be a poor prognostic biomarker of gastric cancer.
