摘要 目的探讨IL-4、IL-10对肾缺血再灌注损伤(IRI)大鼠血清TNF-α、Cr变化的影响及意义。方法将30只雌性SD大鼠随机分为假手术组(Sham组)、手术+0.9%氯化钠注射液对照组(NaCl组)、手术+IL-4组(IL-4组)、手术+IL-10组(IL-10组)、手术+IL-4+IL-10组(IL-4+IL-10组),每组6只。用无损伤动脉夹钳夹大鼠双侧肾蒂45min制成肾IRI动物模型,钳夹松开时,各组迅速注射相应的IL或0.9%氯化钠注射液,12h后取血。在油镜下观察并计数中性白细胞浸润数;用ELISA法检测TNF-α以及Cr水平。结果IL-4及IL-10在降低血清TNF-α。水平上有一定的意义,但均不能降低Cr水平。而联合运用IL-4及IL-10既能降低血清TNF-α水平,也能降低Cr水平。结论联合应用IL-4及IL-10可改善肾功能,其作用机制可能与中性白细胞浸润受抑制及血清中TNF-α减少有关,提示联合应用IL-4及IL-10对肾IRI治疗可能有潜在的价值。 Objective To investigate the effect of IL-4 or / and IL-10 on the serum TNF-α and Cr levels in rat model of renal isehemic-reperfusion injury(IRI). Methods The IRI model was established by clamping bilateral renal pedicles for 45min in 30 female Sprague-Dawley rats. When the clamp was loosed, relevant agents was injected. The animals were divided into five groups (n=6 in each group): sham-operation group (Sham group); operatian+0.9%Nacl control (NaCl group); operation+IL-4(IL-4 group); operation+IL- 10(IL- 10group);operatian+IL-4+IL- 10(IL-4+IL- 10group). Neutrophilie leukocyte infiltration was observed by histopathology examination; Serum TNF-ct and Ser levels were measured by ELISA method. Results IL-4 or IL-10 alone could reduce serum TNF-α level, bur had uo protective effect on renal function. The combination of IL-4 and IL-10 was more effective in redncing TNF- α level and prevented kidney from functional damage. ConcIusion The result indicate that the combination of IL-4 and IL- l0 may prevent kidney from renal ischemie-reperfusion injury.
