细胞周期素E P27^(KIP1)多参数测定在胃癌早期诊断中的应用

摘要 目的探讨细胞周期素E(cyclinE)、P27KIP1的表达在胃癌早期诊断中的价值。方法本研究共选取34例胃癌、10例癌前病变及10例正常胃黏膜,机械法或胶原酶和透明质酸酶混合消化法制备单细胞悬液。异硫氰酸荧光素(FITC)标记的二抗和藻红蛋白(PE)标记的单抗经多参数流式细胞术的方法分别检测cyclinE、P27KIP1的表达。结果胃癌cyclinE和P27KIP1的表达率分别为(32±15)%、(6±5)%,与正常胃黏膜及癌前病变相比较,差异有统计学意义(P<0.01)。结论CyclinE过表达和P27KIP1低表达使G1/S转换点调节失控,与胃癌的发生、进展密切相关。二者的联合测定有利于胃癌的早期诊断。 Objective To explore the expression quantity of cyclinE, p27^kip1, and their application in early diagnosis of gastric carcintmaa. Methods There were 34 patients with gastric cacinoma, 10 patients with pre-carcinoma lesions, and 10 normal gastric muocosa being studied. To make clinic samples into single cell suspensions, mechanical digestion or mixed enzyme digestion of collagenase type Ⅰ and hyaluronidase was used. After incubated with FITC-conjugated goat-anti-mouse monoclonal antibody to cycinE and PE-P27^kip1 Mab, the cultures were examined by flow cytometry with multiple parameters for their expression quantity. Results The cyclinE and p27kip1 expression rate in gastric cancer was (32 ± 15)%, (6 ± 5)% respectively, having considerable difference compared with that of control group (P〈 0.01 ). Conclusion The over expression of cyclinE and low or no expression of p27^kipl during G1/S transition may disturb cell cycle, and be relate to carcinogenesis and progression of gastric carcinoma. The detection of both cyclinE and p27^kipl may apply to early diagnosis in gastric carcinoma.
