补肝养髓方对自发老年性痴呆模型氧自由基损伤的定性定量研究 被引量:7

摘要 目的:观察补肝养髓方对自发老年性痴呆模型学习记忆功能及氧自由基代谢的影响。方法:昆明种自发老年性痴呆(记忆障碍)小鼠,随机分为4组,即空白对照组、西药对照组、补肝养髓方小剂量组、补肝养髓方大剂量组、另设老年学习记忆正常组,分别给药,连续60天。用跳台实验检测学习记忆成绩;脑组织冰冻切片,免疫组织化学法检测大脑皮质及海马SOD活性;海马CA1区脑组织超薄切片,透射电镜观察神经元细胞脂褐素;全自动显微图象分析系统定量检测SOD及脂褐素。结果:补肝养髓方能显著改善自发老年性痴呆模型小鼠的学习记忆障碍(P<0.05-0.01),并明显增强中枢大脑皮质和海马SOD活性(P<0.05-0.01),降低其神经元细胞脂褐素含量(P<0.05-0.01),且其作用呈现出一定的量效关系。结论:补肝养髓方方可明显改善自发老年性痴呆模型的学习记忆障碍,这一作用可能与增强其大脑皮质和海马SOD活性,拮抗氧自由基损害有关。 Objective:To observe the effects of bugan yangsui formula on the ability of learning and memory and synaptic plasticity of neuron in spontaneous aged dementia mice.Methods:Spontaneous aged dementia mice were screened out from 21 months old Kunming mice and then were divided randomly into 4 groups:control,western medicine,lower dosage bugan yangsui and higher dosage bugan yangsui formula groups,in addition,normal aged mice group was also set.Using step-down test to estimate the ability of learning and memory;using transmission electron microscopic examination and automatic micro-image analysis system to determine the parameters of synaptic plasticity in the brain tissue of hippocampal CA1 region.Results:Bugan yangsui formula could improve the disturbance of learning and memory in spontaneous aged dementia mice remarkably(P<0.05~0.01),could increase synaptic numerical density and surface density in molecular layer of hippocampal CA1 region(P<0.05~0.01).All these effects had a dose-response relationship.Conclusion:Bugan yangsui formula could improve the disturbance of learning and memory in spontaneous aged dementia mice significantly,this effect might be related to the ability of bugan yangsui formula accelerating the regeneration of synapse,improving synaptic plasticity of brain.
