高浓度葡萄糖对3T3-L1脂肪细胞脂联素分泌的影响 被引量:2

摘要 目的:探讨高浓度葡萄糖对3T3-L1脂肪细胞脂联素分泌的影响。方法:3T3-L1脂肪细胞诱导分化后分为4组,处理如下,(1)5.5mmol/L葡萄糖;(2)5.5mmol/L葡萄糖+50μmol/LPD98059;(3)17.5mmol/L葡萄糖;(4)17.5mmol/L葡萄糖+50μmol/LPD98059。48h后半定量RT-PCR法检测3T3-L1脂肪细胞过氧化物酶体增殖物激活受体γ(PPAR-γ)mRNA的表达,ELISA法检测培养液中脂联素浓度。结果:(1)高浓度葡萄糖可以降低PPAR-γmRNA的表达、减少3T3-L1脂肪细胞脂联素的分泌;(2)PD98059能够部分逆转由高糖引起的PPAR-γmRNA表达降低及脂联素分泌的下降。结论:高浓度葡萄糖通过激活细胞丝裂原活化蛋白激酶信号传导通路降低PPAR-γmRNA的表达,同时减少3T3-L1脂肪细胞脂联素的分泌。
