流产大鼠下丘脑TNF-α的分布及变化 被引量:1

摘要 为了探讨流产大鼠下丘脑核团肿瘤坏死因子α(tumor necrosis factor alpha,TNF-α)的分布及变化,将12只孕SD大鼠随机分为流产组和妊娠组。流产组在妊娠第10,11天灌服米非司酮(RU486)3.75 mg/kg,妊娠组在妊娠第10,11天灌服等量生理盐水,所有大鼠均于妊娠第13天灌注取材。采用免疫组织化学SP法,对各组大鼠下丘脑TNF-α阳性细胞进行观察,探讨TNF-α在流产大鼠下丘脑的分布及变化。结果表明,妊娠组和流产组大鼠下丘脑视前大细胞核、视上核、室旁核、下丘脑室周核、弓状核均有TNF-α阳性细胞;与妊娠组相比,流产组大鼠各核团阳性细胞数量均极显著增加(P<0.01),灰度值均降低,除视前大细胞核外,均有极显著差异(P<0.01)。除以上核团外,流产组大鼠视交叉上核也有较多TNF-α阳性细胞。结果表明流产大鼠下丘脑核团TNF-α分布及变化较妊娠大鼠增强。 In order to investigate the location and variation of TNF-α in hypothalamus of abortion rats,twelve pregnant SD rats were divided into 2 groups randomly.One was abortion group,and the other was pregnancy group.Rats in abortion group were given RU486 3.75 mg/kg at 10th,and 11th pregnant day,while pregnancy group were only treated with 0.9% NaCl.All rats were affused at 13 th pregnant day.In order to study the location and variation of tumor necrosis factorα(TNF-α) in the hypothalamus of abortion rats,immunochemistry SP was used to detect the positive TNF-α cells.The result were as follows:TNF-α positive cells could be found in nucleus pre-opticus magno cellularis(poma),nucleus supra-opticus(so),nucleus periventricularis hypothalami(hpv),nucleus arcuatus hypothalami(ar) and nucleus paraventricularis hypothalami(pvn) in both two groups;compared with pregnancy group,amounts of TNF-α cells in above nucleuses of abortion group obviously increased,while gray scales were significantly decreased except poma.Besides,nucleus pre-opticus suprachiasmaticus(posc) also expressed TNF-α in abortion group.The results indicated that the expression of TNF-α in hypothalamus of abortion rats would increase.
