摘要
目的:观察铜负荷大鼠肝脏Fas和FasL的表达情况,探讨细胞凋亡在铜损伤肝脏中的意义。方法:建立铜负荷大鼠模型,TUNEL法观察铜负荷大鼠肝脏凋亡改变,免疫组化法测定肝组织Fas和FasL的表达。结果:凋亡指数(AI)随铜负荷时间上升,从正常对照组的(2.23±1.34)%上升到铜负荷模型组的(15.78±2.42)%(P<0.01);与正常对照组相比较,铜负荷组Fas和FasL在肝细胞中的表达显著增强(P<0.01)。结论:过多的铜可以诱导肝脏的凋亡,Fas/FasL系统介导了铜负荷大鼠肝细胞的凋亡。
Objective: To investigate the expressions of Fas and FasLLigand in the liver of copper-overload rats and their significance in liver injury with over copper.Methods:Models of copper-overloading rats were established;TUNEL assay were used to detect apoptosis in liver;The expressions of Fas and FasL-Ligand were detected by immunohistochemistry.Results:The apoptosis index(AI) displayed higher in model rats(15.78±2.42)% than that in normal rats(2.23±1.34)%,and kept increasing with copper overloading time;Compared with control group,the expressions of Fas and FasL-Ligand in model groups were significantly increased(P〈0.01).Conclusion:Over copper induced apoptosis in liver;Fas/FasL system mediated apoptosis of liver in copper-overload rats.
