黑大豆种皮花色苷提取物抗兔动脉粥样硬化作用 被引量:10

THE ANTI-ATHEROGENIC EFFECTS OF BLACK SOYBEAN COAT ANTHOCYANIN EXTRACTS IN RABBITS

摘要 目的研究黑大豆种皮花色苷提取物(black soybean coat anthocyanin extract,BSCAE)对高胆固醇饲料诱导的兔动脉粥样硬化(AS)形成的影响。方法:以成年雄性新西兰大白兔为实验动物,随机分为4组:正常对照组,模型对照组,以及低、高剂量BSCAE组,每组12只。除正常对照组外,其余三组前10w均用高胆固醇饲料喂养。实验组灌胃BSCAE(400,800mg/kgbw),对照组灌胃生理盐水。18w后观察主动脉AS斑块形成情况,测定血脂、肝脏胆固醇含量及血清和肝脏的抗氧化酶活性和脂质过氧化产物丙二醛(MDA)含量。结果:BSCAE抑制了主动脉AS斑块的发展,同时血清胆固醇,低密度脂蛋白胆固醇和载脂蛋白B水平降低。血清和肝脏中MDA生成下降,肝脏超氧化物岐化酶活性增加,800mg/kgbwBSCAE还可减少肝脏中胆固醇含量,提高血清和肝脏中谷胱甘肽过氧化物酶活性。结论:摄入一定剂量的BSCAE可减轻高胆固醇饲料诱导的兔主动脉AS斑块的形成,改善血脂代谢和抗氧化作用是其可能的作用机制。 Objective: To investigate the potential anti-atherogenic effects of black soybean anthocyanin extracts (BSCAE) on rabbits fed with high cholesterol diets. Method: Forty-eight male adult New Zealand white rabbits were randomly divided into four groups: normal control group, model control group and two experimental groups respectively. Except normal control group, the other groups were fed with high-cholesterol diets in previous 10 w. Rabbits were given different doses of BSCAE (400,800 mg/kg bw) in experimental groups or 15ml normal saline in control groups ig every day for 18 w. Then, all animals were killed and serum and liver were collected for determination of lipid profile, malondialdehyde (MDA) content and activity of antioxidative enzymes. Aorta were collected for evaluation of atherosclerotic (AS) lesion grossly by Sudan Ⅲ or histopathologically by HE staining. Results: The gross and histopathological findings confirmed that feeding BSCAE to rabbits restrained the progression of AS lesions in the aorta. Different doses of BSCAE reduced the profile of total cholesterol, low density lipoprotein and apoprotein B as compared to model group significantly. BSCAE decreased MDA both in serum and liver and increased the activity of superoxide dismutase (SOD) in liver, In addition, the elevated liver cholesterol contents and the activities of serum and liver glutathione peroxidase were siginificantly lowered in rabbits fed with 800 mg/kg bw BSCAE than model group. Conclusion: BSCAE can restrain the progression of AS lesion, the potential mechanism for which is the antioxidative and hypocholesterolemic effiects of BSCAE.
