摘要
目的:建立大脑中动脉栓塞(MCAO)模型,观察大豆异黄酮(ISO)对脑缺血大鼠的保护作用。方法:健康成年SD大鼠随机分成6组:ISO低、中、高剂量组、VE组、模型组、假手术组。4w后前5组做MCAO模型,分别于造模后1h和6h磁共振检测抗氧化指标,用透射电镜进行神经细胞形态学观察。结果:与模型组相比,各ISO组大鼠血清SOD和GSH-PX活性、6h各兴趣区rADC、rDCavg、rFA值较高;血清MDA含量较低,差别均有统计学意义。ISO剂量与血清SOD和GSH-PX活性、6h各兴趣区rADC、rDCavg、rFA均呈正相关;而与血清MDA含量呈负相关。形态学观察显示,ISO高剂量组神经元细胞结构良好。结论:ISO可通过提高机体抗氧化能力对缺血性大脑损伤起到一定的保护作用,ISO低、中剂量组与VE组作用相近。
Objective: To study the protective effect of soybean isoflavone (ISO) against focal cerebral ischemia induced by middle cerebral artery occlusion (MCAO) in rats. Method: SD rats were randomly assigned into six groups: ISO-L, ISO-M ,ISO-H, VE, model and sham. ISO and VE were provided for 4 w. Then the left middle artery occlusion was induced as model in the former 5 groups. Ischemic penumbra was observed by MRI in lh and 6h after operation. The antioxidative indices were measured, and the morphological changes of neuron were observed by transmission electron microscope. Results: The serum SOD and GSH-PX activities and rADC, rDCavg, rFA in 6h in interesting areas in ISO groups were, significantly higher than those in model, and positively correlated with the dose of ISO. The serum MDA in ISO groups were significantly lower than those in model and negatively correlated with dose of ISO. The morphological study showed the configuration of neuron in ISO-H group was good. Conclusion: ISO could protect cerebral ischemia through enhancing the activities of antioxidative system. The effects in group ISO-L, ISO-M were similar to group VE.
