摘要
目的:为探讨CTGF在糖尿病肾病发生中的作用,寻找有效的干预药物,观察绞股蓝总皂苷对糖尿病肾病大鼠肾脏CTGF表达的影响。方法:采用肾脏病理形态以及免疫组织化学检查对糖尿病肾病模型大鼠肾脏中CTGF mRNA含量的测定,分析绞股蓝皂苷改善肾功能的机制。结果:病理切片示各造模组大鼠肾小球系膜区细胞外基质呈不同程度的积聚。和正常大鼠(F组)相比,各组CTGF mRNA含量均增高;各给药组(A、B、C、D组)CTGF mRNA的蛋白表达较模型组(E组)降低;D组对CTGFmRNA蛋白表达的降低作用与B组相似。结论:绞股蓝皂苷能明显抑制实验性糖尿病肾病大鼠CTGF表达,从而减少肾小球系膜区ECM的积聚。
Objective: To further investigate the function of CTGFmRNA in DN rat′s during the occurance of DN,and to observe the influence of Gypenoside on CTGF in early diabetic nephropathy. Methods : The content of CTGF in DN rat was measured by pathomorphology and immunohistochemistry, and the mechanism of Gypenosides to improve the renal function was analyzed. Results: Pathological section show the ECM was accumulated at different degree in the modal groups, and comparing to the normal rats (Group F) ,the expression of CTGFin DN rats was increased, and the expres- sion in treatmem groups( Group A ,B, C, D) was lower than the model group. The res;alt in group D was similar to group B. Conclusion: Gypenosides can inhibit the high expression of CTGFmRNA in DN rats, decrease the accumulation of ECM in glomcrulus mesangial region.
