RP-HPLC测定六味木香散中胡椒碱含量

Determination of piperine in Liuwei Muxiang San by RP HPLC

目的：建立六味木香散中胡椒碱含量测定方法。方法：反相液相色谱法。采用ＳｈｉｍＰａｃｋＣＬＣＯＤＳ（１５ｃｍ×６ｍｍ）分析柱；ＯＤＳ保护柱；流动相：甲醇水（６７∶３３）；流速：１．０ｍｌ·ｍｉｎ－１；色谱柱温度３５℃；检测波长：３４３ｎｍ；进样体积１０μｌ；外标法计算含量。结果：理论板数按胡椒碱峰计算为３６００；分离度为２．５；胡椒碱浓度在３～４８μｇ·ｍｌ－１范围与色谱峰面积呈线性关系，ｒ＝０．９９９９；回收率及ＲＳＤ分别是９８．６％和２．１％。结论：方法简便准确，可用于六味木香散中胡椒碱含量测定。

OBJECTIVE: To establish a quantitative method of piperine in Liuwei Muxing San(Chinese petent medicine). METHODS:Piperine in Liuwei Muxiang San was determinated by RP HPLC. Chromatographic conditions included column ODS C 18 , column temperature, 35℃; detector UV, 343 nm; mobile phase, methanol water(67∶33); flow rate, 1 ml·min -1 ; injecting volume, 10 μl; external standard. RESULTS: The number of theoretical plates calculated by piperine peak was 3 600. The resolution was 2.5. The standard curve was linear in the concentration range of 3～48 μg·ml -1 ,and correlation coefficient was 0.999 9. The average recovery and the relative standard deviation were 98.6% and 2.1%,respectivily. CONCLUSION: The method is simple, accurate, and it is satisfactory for determination of piperine in Liuwei Muxiang San.
