溃疡性结肠炎患者PBMC体外白细胞介素12诱生水平研究 被引量:5

摘要 目的探讨白细胞介素12(IL-12)在溃疡性结肠炎(UC)中的表达水平。方法以外周血单个核细胞(PBMC)体外培养诱生IL-12。以放射免疫法分析检测IL-12的水平,将健康人(HC)与溃结患者进行比较研究。其中HC组55例,UC组45例(静止期与轻、中、重组例数分别为9、18、11、7)。结果UC组与HC组IL-12水平分别为(301.22±82.01)ng/L和(110.00±53.95)ng/L,P<0.001。UC组静止期与轻、中、重三组比较IL-12水平依次为:(142.01±47.05)ng/L、(248.73±60.63)ng/L、(295.08±80.40)ng/L、(335.20±119.95)ng/L,P值分别小于0.02、0.02、0.001。而静止期与HC组相比P>0.05。按病变累及范围,全结肠组与左半结肠组IL-12水平分别为:(312.83±106.25)ng/L、(264.85±67.97)ng/L(P>0.05)。结论UC活动期患者IL-12活性明显高于对照与静止期患者,且随病情加重呈上升趋势。IL-12活性与病变累及范围无关。 Objective To study the potency of IL-12 production in peripheral blood mononuclear cells(PBMC) between 45 patients with inactive and active stage (including mild, moderate and severe)ulcerative colitis(UC)and age and sex-matched 55 healthy volunteers. Methods PBMC were stimulated with PHA for 72hrs to induce IL-12 production. IL-12 content in the culture medium was assayed by using RIA. Results IL-12 production was found to have significantly increased in PBMC from active UC. There was an increasing tendency compared with the severe degree of the disease, but no significant difference was found among the location of the injury. Conclusion The potency of IL-12 production may indicate some important role of IL-12 in the pathogenesis of UC and also the potency of IL-12 production in PBMC can be an indicator of the activity of UC. Inhibition of IL-12 might be future therapeutic targets in IBD.
