摘要
目的观察通心络超微粉和褪黑素对实验性糖尿病大鼠血浆、肾组织血管紧张素(Ang)水平的联合干预作用。方法采用2×2析因试验设计,将28只雄性SD糖尿病大鼠随机平均分配到糖尿病未干预组(DM组)、通心络超微粉干预组(TXL组)、褪黑素干预组(MEL组)、通心络超微粉联合褪黑素干预组(TXL+MEL组)。分别连续给药12周后,取血浆、肾组织,采用放射免疫分析法测定血浆和肾组织AngI、AngII水平。结果各组血浆和肾组织AngI、AngII水平以TXL+MEL组最低,DM组最高,TXL组或MEL组居中。结果显示单独用TXL或MEL均能降低肾组织、血浆Ang水平,联合干预对降低肾组织AngII、血浆AngI水平存在交互作用,对降低肾组织AngI、血浆AngII水平无交互作用;交互作用精细比较时仍显示对肾组织AngII的调节作用,联合干预与单独干预的作用有差异。结论(1)通心络超微粉和褪黑素对降低糖尿病大鼠肾组织AngII水平具有良好的协同作用;(2)单独应用TXL或MEL都能有效降低血浆和肾组织Ang水平;(3)析因设计资料的正确分析有助于信息的充分利用。
Objective To observe the complex intervention effects of Tongxinluo superfine powder and melatonin on angiotensin(Ang) levels in plasma and kidney in rats with diabetes mellitus(DM). Methods Using 2 × 2 factorial design,28 male SD experimental diabetic rats were randomly divided into four gorups, non-intervention DM group ( DM group), Tongxinluo (TXL) superfine powder intervention group ( TXL group), melatonin-intervention group(MEL group) and complex intervention group (TXL+ MEL group). Meantime 7 normal SD rats were included in controlgroup. After 12-day administrafion,the levels of Ang Ⅰ and Ang Ⅱin plasma and kidney of rats were detected. Results The levels of Ang Ⅰand Ⅱin plasma and kidney of TXL + MEL group were the lowest among four groups, while which in DM group were the highest, the levels of TXL group and MEL group were between the two groups. According to the results of factorial design ANOVA, it was seen that both only TXL and only MEL were effective in lowering the levels of Ang Ⅰand Ⅱin plasma and kidney, there was a interaction in complex intervention in lowering the levels of Ang Ⅱin kidney and Ang Ⅰin plasma,however, there was no interaction in lowering the levels of Ang Ⅰin kidney and Ang Ⅱin plasma. There was a significant difference in the intervention effects between complex intervention and only intervention. Conclusion There is a close synergistic effect in TXL superfine powder combined with melatonin on reducing the levels of Ang Ⅱin kidney of DM rats. Only administration with TXL superfine powder or melatonin could reduce effectively the levels of angiotensin in plasma and kidney. The correct analysis for factorial design data is helpful to the fully application of information.
