外阴阴道念珠菌病小鼠模型阴道组织PGE_2的测定及消炎痛对其治疗的研究 被引量:4

摘要 目的:探讨雌激素诱发白念珠菌性阴道炎的发病机制,观察阴道内应用消炎痛对白念珠菌性阴道炎的影响并分析其作用机制。方法:采用雌激素化小鼠白念珠菌性阴道炎模型(A)、另设单一白念珠菌感染组(B)、单一雌激素化组(C)、模型用药(消炎痛)组(D)和空白对照组(E)。在接种后4、7、14天取小鼠阴道灌洗液并摘取阴道组织,动态观察灌洗液中白念珠菌菌丝生长情况。采用酶联免疫吸附(ELISA)法检测小鼠阴道组织前列腺素E2(PGE2)水平。结果:与E组比较,A组和C组小鼠阴道组织PGE2水平均增高;与B组及D组比较,A组阴道灌洗液中见更多菌丝生长,且阴道组织PGE2水平增高。结论:雌激素可诱导小鼠阴道组织产生PGE2,与白念珠菌菌丝生成和阴道炎形成及持续感染有关;消炎痛能抑制小鼠阴道内白念珠菌菌丝形成和PGE2合成,阴道内给予消炎痛有助于治疗白念珠菌性阴道炎。 Objective: To investigate the pathogenesy of vulvovaginal candidiasis (VVC) induced by estrogen, and to observe the effect of indomethacin on a murine model of VVC and analyze the mechanism of action. Methods: The estrogen - treated murine model of VVC (A) was set up. The infected but non - estrogen - treated mice ( B), estrogen - treated but uninfected mice ( C), indomethacin - treated mice (D) and naive mice (E) also were established. The vaginal lavage fluid and vaginal tissues were obtained at day 4, 7, 14 after inoculation and the hypha were observed. Enzyme - linked immunosorbent assay (ELISA) was applied to determine prostaglandin E2 (PGE2) levels in the tissues. Results: The levels of PGE2 in A and C were higher than those in E. More hypha and significantly higher levels of PGE2 were observed in vaginal tissues in A than those in B and D. Conclusion: Estrogen may up - regulate the PGE2 levels in murine vaginal tissues, which may be associated with the hypha formation and the onset of VVC. Indomethacin can inhibit the hypha formation and PGE2 production in vaginal tissues. The administration of indomethacin intravaginally should be helpful to the treatment of VVC.
