摘要
应用二种中性盐溴化红和氯化钙作为溶解蚕丝丝素的试剂,制成了二种固定化葡萄糖氧化酶丝素膜。经酶比色法和红外分光光度法分析,结果表明这二种丝素膜都是良好的固定化酶的生物材料。葡萄糖氧化酶经这些丝素膜固定以后对热和pH稳定性得到明显改良。这些酶膜性能稳定,具有高的活性得率,能长期存放。以这些固定化葡萄糖氧化酶丝素膜和氧电极为基础,研制的流动注射分析式电流型葡萄糖生物传感器性能较稳定,具有较宽的葡萄糖线性响应范围(0.5~15.0mmol/L),相关系数为0.999,可以反复测定葡萄糖一千次以上。
Glucose oxidase(GOD) was immobilised in two silk fibroin membranes prepared by using two neutral salts LiDr and CaCli as a reagent dissolving fibrion derived from Bombyx mori cocoon. Two fibrion membranes were demonstrated to be good enzyme -immobilized materials by rneans of enzymatic-coloranertric method and infrared spec-trophotometry. After GOD was immobilised in both fibrion membranes, its stability to pH and heat was greatly improved. These enzyme membranes are of high recoveries of enzyme activity and could be stored for long term. Eased on these GOD-immobilized fibroin membranes and oxygen electrode, an amperomctric sensor in flow injection analysis was developed for monitoring glucose solution. The regression coefficients for the linear regions of the biosensor(.up to 15. 0 mmol/L)are 0.999. These GOD membranes in the biosensor can be rtsed repeatedly for glucose determination over 1000 times.
