摘要
目的研究神经生长因子对体外培养的人牙周膜细胞增殖和分化作用的影响。方法采用四唑盐比色法(MTT)和碱性磷酸酶(ALP)活性检测法,测定不同浓度(0.1U/ml、1U/ml、10U/ml、100U/ml)的神经生长因子对体外培养的第5-6代人牙周膜细胞增殖及碱性磷酸酶活性的作用。结果与对照组(2%FBS的DMEM培养液)相比,1,10,100U/ml的神经生长因子都可促进人牙周膜细胞的增殖,但10U/ml作用最显著;100U/ml的神经生长因子可显著促进碱性磷酸酶的活性(P<0.01)。结论不同浓度的神经生长因子可显著促进人牙周膜细胞的增殖与分化。
Objective To evaluate the effects of nerve growth factor(NGF) on proliferation and differentiation of human periodontal ligament cells(hPDLCs) in vitro. Methods hPDLCs were cultured by explant separation method. The fifth to sixth generation hPDLCs were cultured with 0.1 U/ml, 1 U/ml, 10 U/ml, 100 U/ml NGF for 24 h in experiment groups. Colorimetric MTT metlnod and enzyme dynamics method were used to observe the proliferation of hPDLCs and alkaline phosphatase(ALP) activities. Results Compared with control group(cultured with 2% FBS-DMEM), the cell proliferation significantly increased in 3 experiment groups (cultured with 1, 10, 100 U/ml NGF-DMEM), highest in 10 U/ml NGF-DMEM group (P〈0.05). ALP activities also significantly increased in 3 experiment groups (cultured with 1, 10, 100 U/ml NGF-DMEM), highest in 100 U/ml NGF group(P〈 0.01). Conclusion Different concentrations of NGF could promote proliferation and differentiation of hPDLCs in vitro.
