单纯性肥胖及2型糖尿病儿童胰岛素受体底物的研究

摘要 目的探讨单纯性肥胖,2型糖尿病儿童胰岛素受体底物-1,2的表达。方法对20例单纯性肥胖儿童,20例2型糖尿病儿童及20例对照组儿童,采用细胞免疫技术进行白细胞染色,运用图像分析软件分析,利用光密度值,定量比较白细胞中胰岛素受体底物-1,2的含量变化。结果与对照组相比,肥胖组,2型糖尿病组胰岛素受体底物-1表达减少(P<0.05),而胰岛素受体底物-2表达均无统计学差异(P>0.05)。结论肥胖儿童和2型糖尿病儿童的胰岛素受体底物-1表达下降可能是其出现高胰岛素血症的一个原因,但胰岛素受体底物-2可能不会引起胰岛素血症。 Objective To explore the expression of insulin receptor substrate(IRS) in children with obesity and type 2 diabetes. Methods White blood cells were measured and assessed by immnunocytochemistry and computer imaging analysis system in 20 children with obesity, 20 with type 2 diabetes and 20 controls. Expression of IRS-1,2 was quantitatively determined with optical density values. Results Compared with control children, the expression of IRS-1 significantly decreased in obesity, type 2 diabetes children (P〈0.05), and the expression of IRS-2 increased, but there was no significant difference between 3 groups (P〉0.05). Conclusion The downregulation of IRS-1 expression might induce hyperinsulinemia in children with obesity and type 2 diabetes, but IRS-2 might not induce hyperinsulinemia.
