多重聚合酶链反应法检测质粒介导AmpC酶的优化及应用

摘要 目的建立快速检测质粒介导AmpC酶的分子生物学方法。方法采用多重聚合酶链反应(PCR)法并优化反应体系,检测21株头孢西丁耐药大肠杆菌质粒型AmpC酶基因型。结果反应体系中dNTP和引物浓度、循环参数中退火温度对多重PCR的扩增效果有影响;反应体系中缓冲液、Mg2+浓度、反应体积、循环参数中延伸温度、循环次数对多重PCR的扩增效果影响很小;21株头孢西丁耐药大肠杆菌中8株质粒AmpC酶基因型阳性,其中DHA基因型5株,ACC基因型3株。结论多重PCR技术检测质粒介导AmpC酶效果较好,具有可行性。为临床提供了简单易行、快捷有效、敏感、特异性高的方法。
