摘要
用高效液相色谱电化学检测法测定血浆中丙咪嗪及其代谢物浓度。以1%三乙胺磷酸缓冲液(pH3.0):甲醇:四氢呋喃(60:30:10,v/v)为流动相,在电化学检测器0.7v的工作电压下进行检测。丙咪嗪、去甲丙咪嗪、2-羟基丙咪嗪和2-羟基去甲丙咪嗪的最低检测浓度分别为0.5,0.5,0.6和0.8μg/L血浆,平均萃取回收率分别为86.9%,88.4%,82.5%和78.8%,日内CV%分别小于3.96,4.72,6.05和6.64,日间CV%分别小于5.63,5.33,4.95和6.24。方法简便、灵敏,可充分满足临床药理研究和治疗药物监测工作的要求。
A high-performance liquid chromatography with electrochemical detection was developed for the determination of imipramine and its metabolites in plasma. The mobile phase consisted of 1% triethylamine (pH 3.0,adjusted with phosphoric acid)-methanol-tetrahydrofuran (60:30:10, V/V)and electrochemical detection at 0.7 V.The detection limit for imipramine, desipramine, 2-hydroxyimipramine and 2-hydroxydesipramine were 0.5, 0.5, 0.6 and 0.8 μg/L, respectively. The mean recovery were 86.9%, 88.4%, 82.5% and 78.8%, respectively.The coefficient of variation(CV%) of within-day were less than 3.96, 4.72, 6.05 and 6.64, respectively. The between-day CV% were less than 5.63, 5.33, 4.95 and 6.24, respectively. The method is simple and sensitive enough to be used for clinical pharmacological research and therapeutic drug monitoring.
