中药灌洗液抗小鼠肉芽肿增生的实验研究

Experiment Study of Chinese Herbal Irrigative Solution on Nasal Mucosa of Post-surgery Nasosinusitis Mice

目的:研究鼻窦炎术后中药灌洗液局部运用对试验性小鼠慢性肉芽肿增生的抑制作用。方法:①将18只小鼠随机分为生理盐水对照组和中药浓缩液实验组,两组分别用0.9%生理盐水和中药浓缩液(灌洗液浓缩35倍)滴鼻,每天8次,连续给药7d后,肉眼观察两组小鼠的全身一般情况。颈椎断裂处死小鼠后肉眼观察鼻黏膜变化。②将40只小鼠随机分为模型组、中药灌洗液高剂量组、中药灌洗液低剂量组和醋酸氟轻松对照组,每组10只。鼠背酒精消毒后于鼠背中线处皮下注射2%琼脂溶液0.4mL,分别用0.9%生理盐水、高剂量中药灌洗液、低剂量中药灌洗液以及醋酸氟轻松软膏涂抹鼠背,连续7d,每日2次。末次给药24h后,颈椎断裂处死,小心剥离出肉芽肿琼脂块,称取湿重,观察对比各组小鼠肉芽肿琼脂块湿重。结果:①各组小鼠一般情况良好、生理盐水对照组和中药浓缩液治疗组小鼠无明显差异。②中药灌洗液高剂量组、醋酸氟轻松对照组与模型组比较,肉芽肿琼脂块湿重有显著差异(P<0.05)。结论:中药灌洗液对鼻腔黏膜无局部刺激作用,能够抑制慢性肉芽肿的增生。

Objective： To investigate the effect of Chinese herbal irrigative solution on nasal mucosa of mice after nasosinusitis surgery. Methods： （1）Acute toxicity test：18 mice were randomly divided into saline control group and concentrated Chinese herbal solution treatment group,then saline and concentrated Chinese herbal solution were respectively dripped into nasal cavities of mice, the general condition of mice were observed and the alterations of nasal mucosa were observed after the mice were sacrificed by cervical dislocation. （2）Granuloma model rats were induced in 40 mice by subcutaneous injection of agar, then they were randomly divided into blank group, fluocinonide control group, Chinese herbal irrigative solution group 1 （low dosage）and Chinese herbal irrigative solution group 2（high dosage）, weight of granuloma were measured. Results： （1）The difference of general condition was not obvious between saline control group and concentrated Chinese herbal solution treatment group. （2）Compared with the model group, the weight of granuloma were decreased significantly in high dosage of Chinese herbal irrigative solution group and fluocinonide control group, but there were no significant difference between high dosage of Chinese herbal irrigative solution group and fluocinonide control group. Conclusion： Chinese herb can decrease the weight of granuloma tissue and has no stimulative effect on nasal mucosa.
