摘要
本实验检测了一种恶性疟原虫多价重组候补疫苗在小鼠体内引起的体液免疫和细胞免疫反应。该重组疫苗包含了MSA1、MSA2、RESA上的T细胞和/或B细胞刺激位点及IL-1、TT上的T细胞刺激位点;尤其是含有Spf66的序列。经重组疫苗免疫后,BALB/c小鼠和C57BL/6小鼠均产生了明显的抗重组疫苗的抗体,且经重组疫苗免疫的BALB/c小鼠脾细胞在体外经重组疫苗刺激后发生了明显的增殖,并产生了明显的IL-2。以上结果提示该候补疫苗具有免疫原性,且具有一定的T细胞刺激能力和一定的广谱识别能力,其能否有效地刺激人淋巴细胞还需进一步研究。
The cellular and homoral immune responseto a pooyvvlenterecombinant candidate vaccine of Plesmodium falciparam(P.f) were investigated in BALB/c and C57BL/6 mice by studying anti-candidate vaccine antibody, splenocyte proliferation and release of IL-2. This vaccine contains T and/or B Cell epitopes of MSA1、MSA2、RESA of P. f, universal T cell epitopes of interleukin-1(IL-1), Tetanus toxoid(TT) and especially,the sequence of SPf66. Antibody of BALB/c and C57BL/6 mice immunized with candidate vaccine significantly raised antibody to candidate vaccine.The splenocyte of BALB/c mice immunized with candidate vaccine proliferated significantly to candidate vaccine in vitro,it also released significantly IL-2 following stimulation with candidate vaccine. These results suggested that this vaccine should be immunogenic and could stimulate T cell response,and also can be recognized by two MHC haplotypes. It will be further studied if this vaccine is suitbale for person.
