摘要 目的获得人脑源性神经营养因子(hBDNF)基因并进行序列分析,为基因治疗提供参考。方法提取胚胎脑组织基因组mRNA,应用RT-PCR技术获得hBDNF基因序列酶切鉴定,并进行序列测定和分析。结果DNA序列测定的结果与GenBank提供的已知序列(AY054406)比较,所克隆的hBDNF基因从起始密码子ATG到终止密码子TAG全长共744 bp,序列完全相同。结论hBDNF基因序列的获得,为进一步开展面神经损伤的基因治疗积累了资料。 Objective To perform molecular cloning and sequencing of the human brain derived neurotrophic factor (hBDNF) gene for reference of the gene therapy,Methods The human mRNA was extracted from the embryo brain. The hBDNF gene was cloned by RT - PCR, identified by the restriction enzymes, sequenced and analyzed. Results Comparing the cloned hBDNF gene with the GenBank (AY054406) sequence,we demonstrated that both of sequence were from the initiation codon ATG to the termination TAG identieally. The cloned hBDNF gene was 744bp in length. Conclusions Cloning the hBDNF gene has paved the way for further study on genetherapy of facial nerve injury.
