摘要
目的探讨低氧对体外培养的绒毛组织可溶性血管内皮生长因子受体1(sFlT-1)表达的影响。方法孕早期的绒毛组织体外分两组培养:常氧照组和缺氧组。应用RT-PCR方法检测sFlT-1基因mRNA表达水平,应用酶联免疫吸附法(ELISA)检测培养液中sFlT-1蛋白水平。结果与常氧组相比,缺氧组绒毛组织的sFlT-1基因mRNA表达水平增加,sFlT-1蛋白水平也增加。结论缺氧可以诱导体外培养的绒毛组织sFlT-1基因表达,产生sFlT-1增加。缺氧可能是子痫前期胎盘组织sFlT-1高表达的原因。
Objective To investigate the effect of hypoxia on expressions of sFlT-1 in villous explants. Methods The villous explants to be cultured were divided into two groups:control group and hypoxic group. The mRNA expression of sFlT-1 was assayed by reverse transcription-polymerase chain reaction( RT- PCR), and the protein of sFlT-1 was measured by enzyme linked immunosorbent assay (ELISA). Results Compared with the control group, the mRNA expression of sFlT-1 and the level of sFlT-1 protein increased in hypoxie group. Conclusions Hypoxia induces the expression of sFlT-1. Hypoxia may be a cause of high expression of sFlT-1 in preeclamptie placenta.
