食物过敏动物模型中辅助性T淋巴细胞1/辅助性T淋巴细胞2相关细胞因子变化 被引量:8

摘要 目的观察食物过敏动物模型辅助性T淋巴细胞(Th)1/Th2型细胞因子变化,探讨其在食物过敏发生中作用。方法选取110只Balb/c小鼠,用卵清蛋白(OVA)致敏、激发建立食物过敏实验性小鼠模型,设立对照组及致敏组,进行食物过敏原皮肤点刺及小肠病理组织学检测,ELISA检测其血清特异性IgE、IL-4、IFN-γ水平,real-tim e PCR检测其小肠IL-10、TGF-βmRNA表达。结果致敏组小鼠IL-4水平较对照组升高、IFN-γ水平较对照组降低,均有统计学意义(Pa<0.05);其小肠IL-10、TGF-βmRNA表达较对照组降低,有统计学意义(Pa<0.05)。结论食物过敏是以Th2型淋巴细胞占优势的过敏性疾病;Th1/Th2细胞因子失平衡可能参与食物过敏发生,并可能在其中发挥重要作用。 Objective To investigate the function of helper T lymphocyte cell (Th) 1/Th2 cytokine in food allergy development. Methods A total 110 Balb/c mice were randomly divided into 2 groups:control group (40 mice)and food allergy group (70 mice). Food allergy animal models were established by ovalbumin, performed by skin prick test; in positive reaction mice,serum specific IgE,IL-4 and IFN-γ were measured by enzyme linked immuosorbent assay ( ELISA ), and intestinal pathology were performed, and the mRNA expressions of IL - 10, TGF - β in intestinal were measured by real - time PCR assay. Results In food allergy group,the mRNA expression of IL - 10, TGF - β in intestinal decreased (P 〈 0.05 ). Serum IL- 4 increased, while IFN -γ decreased( Pa 〈 0.05 ) , which was significant statistically. Conclusion Food allergy is a disease of predominating by Th2 lymphocyte,and Thl/Th2 cell may play a key role in the pathogenesis of food allergy.
