摘要
目的:检测维A酸干预前后卵巢癌细胞COC2中维A酸受体RARα和RXRαmRNA的表达情况,从分子生物学水平探讨维A酸对卵巢癌细胞的作用。方法:利用实时定量PCR方法检测经不同浓度维A酸干预前后卵巢癌COC2细胞中维A酸受体RARα和RXRαmRNA的表达。结果:维A酸受体RARαmRNA表达在维A酸干预后的卵巢癌细胞株COC2中有所升高,且在一定范围内随维A酸浓度增高而升高;而RXRαmRNA在维A酸干预后表达降低,且在一定范围内随维A酸浓度增高而降低。结论:维A酸干预后,卵巢癌细胞维A酸受体RARα的表达上调,RXRα表达下调,说明维A酸所产生的抗卵巢癌作用可能是通过改变核受体mRNA的表达水平而产生的。
Objective : To detect the expression of retinoic acid receptor RARer and RXRα mRNA in human ovarian cancer cell line COC2 treated with Tretinoin. Methods: Human ovarian cancer cell line COC2 was treated with different concentrations of Tretinoin, and then the expression of retinoic acid receptors RARer and RXRα mRNA was examined using real-time quantitative sion of RARer mRNA was up-regulated and RXRα mRNA was down-regulat PCR. Results : The expresed after treated with Tretinoin in a dose-dependent manner within the range of 1 - 30 μmol/L. Conclusion : Tretinoin induces the regulation of the expression of RARer mRNA and down-regulation of the expression of RXRα mRNA in uphuman ovarian cancer cell line COC2, which serves to explain that its antitumous effect is produced by altering the exoression of retinoic acid receptor mRNA.
