摘要
目的观察急性脑梗死患者外周血单核细胞Toll样受体4(TLR4)的表达及其与TNF-α、IL-6浓度的相关性,初步探讨TLR4在急性脑梗死缺血性炎性损伤中的作用,为临床治疗脑梗死提供新的思路。方法分别运用Tapman实时定量PCR及流式细胞仪法检测39例急性脑梗死患者外周血单核细胞(淋巴细胞、单核细胞)TLR4 mRNA、蛋白的表达,用ELISA法测定血清TNF-α、IL-6的浓度,与35例同期正常对照及32例脑动脉粥样硬化患者比较。结果脑梗死组发病第3天TLR4表达、血清TNF-α、IL-6浓度均显著高于正常对照组和动脉粥样硬化组(P<0.01)。TLR4表达与血清炎性因子TNF-α、IL-6浓度变化呈正相关(P<0.01)。结论研究提示TLR4可能在急性脑梗死缺血性炎症性损伤中具有重要的作用,其机制可能是通过上调TLR4表达促使炎性因子产生分泌来介导脑缺血炎性损伤。
Objective To primarily discuss the role of TLR4 in ischemical inflammation injury of cerebral infarctionion and to provide the new measure to cure cerebral infraction, the expression of Toll-like receptor 4(TLR4) on peripheral-blood mononuclear cells and its correlation with concentration of TNF-α and IL-6 in the patients with acute cerebral infraction were investigated. Methods The expression TLR4 mRNA and protein on peripheral-blood mononuclear cells in 39 cases of acute cerebral infraction were examined by real-time quantitative reverse transcription ploymerse chain reaction (RT-PCR) and flow cytometry (FCM) respective- ly. The content of TNF-α and IL-6 in serum were examined by ELISA. Thirty-five cases of health people and 32 cases of cerebral atherosclerosis were regarded as control groups. Results The expression of TLR4 and the content of TNF-α and IL-6 in serum were significantly higher than those in control groups at 3d after acute cerebral infractionion onset (P〈0.01), and the expression of TLR4 was correlated positively with the concentration of TNF-α and IL-6. Conclusion TLR4 may play a critical role in the mechanism of cerebral inflammation injury, which may be through spuring the excretion of inflammatory factor to arise the cerebral ischemical inflammation injury.
