摘要
目的探讨1,6-二磷酸果糖(FDP)对脑出血后血肿周围脑组织能量代谢产物及神经细胞凋亡的干预作用。方法健康家兔随机分成4组:正常组,脑出血组,治疗组,假手术组。其中脑出血、治疗组、假手术组又分成1、6、12、24、48、72h组。采用二次注血法制备兔自体血脑出血模型。治疗组予果糖二磷酸钠注射液(4.4ml/kg)治疗。高效液相法测定三磷酸腺苷(ATP)含量,TUNEL法测定6、24、72h组血肿周围组织神经细胞凋亡。结果治疗后1、6、12h各组间校正后的ATP含量无显著性差别,24h后治疗组血肿周围组织ATP含量明显增高,并在48、72h保持较高水平,明显高于脑出血组,但仍低于正常组及假手术组,差异均有统计学意义。脑出血后6h有少量凋亡细胞出现,24h明显增多,72h凋亡细胞大量出现,FDP治疗后,各时间点凋亡细胞均较模型组明显减少(P<0.05)。结论FDP可改善血肿周围组织能量代谢,使血肿周围组织凋亡细胞减少。
Objective To investigate the effect of 1, 6-biphosphatofructose on the energy metabolism and apoptosis of perihemotoma region. Methods The rabbits were divided into 4 group: normal group(N), sham group(SH) ,hemorrhage group (ICH) and treatment group (TR). The SH,ICH and TR group were subdivided into 1,6,12,24,48 and 72h group. The model of cerebral hemorrhage in rabbit was made with autologous blood infusion. The content of adenosine triphosphate (ATP) and the change of apoptosis in perihemotoma region in all groups were compared. Results There was no difference in all groups at 1, 6, 12h. But at 24, 48h, 72h ATP content of TR group was higher than that of ICH group whereas lower than that of N and SH groups (P〈0. 05). There were few apoptosis cells at 6h postinfusion, the number of apoptosis cells increased markedly at 24h, and there were a lot of apoptosis cells at 72h. Treating with FDP, the number of apoptosis cells in all time was less than that of ICH group. Conclusion FDP could improve the energy metabolism and ameliorated the apoptosis in the perihemotoma region.
