摘要
目的建立一种用斯达尔样本工作站提取全血中DNA的方法,并评价DNA的纯度和产量。方法使用斯达尔样本工作站提取全血DNA;DNA的纯度和含量用紫外分光光度法测定,并与手工提取方法进行比较。结果斯达尔样本工作站与手工提取的DNA相比,在纯度与得率方面无差异,但更快速、简单。结论本方法可以快速、高效地从大批量全血中提取较高质量的DNA,所得DNA适用于HLA的分型实验。
Objective To estimate a DNA extracting method from whole blood with ML-STAR sample workstation,and to evaluate the quantity and purity of these DNA samples. Methods Genomic DNA was extracted from blood samples by sample workstation. The yield and purity of DNA samples were determined by UV-spectrophotometer and compared with those extracted by traditional method. Results Compared with the traditional method, ML-STAR sample workstation provided a faster and simpler method for blood DNA extraction without decreasing the yield and purity of DNA samples. Conclusion High quality DNA can be extracted rapidly and effectively from whole blood by using ML-STAR sample workstation,and these DNA samples are suitable for HLA typing.
