摘要
目的建立大鼠高眼压视网膜缺血再灌注(IR)模型,观察三七总皂甙(PNS)对大鼠视网膜IR损伤的防护作用和对核转录因子-κB(NF-κB)表达的影响。方法SD大鼠随机分为IR组、IR+PNS组。建立112.5mmHg、60min高眼压视网膜缺血模型,在再灌注后不同时间段处死大鼠取出眼球,光镜观察视网膜损伤后的组织病理学改变,行原位细胞凋亡检测,免疫组织化学法检测NF-κB的表达情况。结果IR组出现视网膜水肿、空泡变性、核固缩等组织病理学改变。IR+PNS组视网膜组织形态学损害明显减轻。原位细胞凋亡检测IR组的凋亡细胞阳性表达明显高于IR+PNS组(P<0.05)。IR组NF-κB的表达明显高于IR+PNS组(P<0.05),IR+PNS组的NF-κB表达滞后。结论PNS可通过抑制NF-кB的活化和减少凋亡而减轻视网膜IR损伤。
Objective Panax Notoginseng Saponins(PNS)is the major effective constitent of Panax notoginseng,and it used extensively in clinic. This study was curried out in the purpose of assessing the effect of the Panax Notoginseng Saponins (PNS) on rat retinal ischemia-reperfusion and NF-κB expression. Methods 112.5 mmHg of pressure was infused into anterio chamber by pressure meter for 60 minutes and then decrease the pressure to normal gradually to create the ischemia-reperfusiol model in 48 Sprague-Dawley rats. PNS was intraperitoneally injected once per day for 6 days in ischemia-reperfusion + PN group( IR + PNS) ,and in control the same amount of normal safine solution was used in ischemia-reperfusion group(IR). Th eyeballs of experimental rats were enucleated and fixed after ischemia-reperfusion at different time intervals for histologica examination of light microscope. Cell apoptosis was evaluated by terminal dUTP nick end labeling(TUNEL) and NF-KB expressiol in retina by immunohistochemistry. Results Histopathological changes were revealed in IR group, including retinal edema vacuolar degeneration etc.. The histopathological damage of retina in IR + PNS group was obviously lighten in comparison with IR group. TUNEL positive cells in IR group was higher than in IR + PNS group at various time points(P 〈0.05). The expression o NF-KB in IR + PNS group was lower than in the IR group(P 〈 0.05). ConcIusion PNS protects retina from damage inducel by IR through lightening apoptosis and restraining NF-KB activation.
