摘要
目的 :探讨多房棘球绦虫 ( Echinococcus multilocularis,E.m.)原头节 18k Da、16k Da抗原 ( E.m.18k Da和 E.m.16k Da)诊断泡球蚴病的应用价值。方法 :应用免疫印迹法检测 2 4例泡球蚴病、55例囊型包虫病、33例囊虫病和 30份健康人血清对 E.m.18k Da和 E.m.16k Da的抗体反应 ,同时用 E.m.2 - EL ISA试剂盒对比检测上述血清对 E.m.2的抗体反应。结果 :在免疫印迹试验中 ,E.m.18k Da和 E.m.16k Da均能被所有 2 4例 ( 10 0 % )泡球蚴病人血清识别 ,与囊型包虫病病人血清交叉反应率分别为 16.4 %和 2 9.1% ,与囊虫病人血清交叉反应率分别为 3.0 %和 33.3%。E.m.2 - EL ISA检测泡球蚴病患者血清抗体阳性率为 95.8% ,与囊型包虫病和囊虫病患者血清交叉反应率分别为 2 3.6%和 33.3%。健康人血清均不与 E.m.18k Da、E.m.16k Da或 E.m.2发生反应。结论 :E.m.18k Da/16k Da,特别是 E.m.18k Da对诊断泡球蚴病有高度敏感性和较强特异性 ,可望成为泡球蚴病免疫诊断的候选抗原。
AIM:To study the usefulness of 18k Da/ 16 k Da antigens in larval Echinococcusmultilo- cularis,designated E.m.18k Da and E.m.16 k Da,for immunodiagnosis of human alveolar echinococcosis.METHODS:Western blot analysis based on the detection of antibody re- sponses against E.m.18k Da and E.m.16 k Da and commercially available E.m.2 - ELISA were evaluated comparatively using2 4sera from patients with alveolar echinococcosis,5 5 sera from patients with cystic echinococcosis,33sera from patients with cysticercosis and30 sera from healthy controls.RESUL TS:All 2 4(10 0 % ) sera from alvrolar echinococcosis pa- tients recognized both E.m.18k Da and E.m.16 k Da in Western blotting,whereas2 3of them were positive in E.m.2 - EL ISA.The cross- reactivity with sera from Cystic echinococ- cosis was 16 .4% for E.m.18k Da,2 9.1% for E.m.16 k Da and 2 3.6 % for E.m.2 ,while the cross- reactivity with sera from Cysticercosis was3.0 % for E.m.18k Da,33.3% for both E.m.16 k Da and E.m.2 .None of the sera from healthy controls showed reactivity with E.m.18k Da,E.m.16 k Da or E.m 2 .CONCL USION:Ourfindings confirmed the high potential of E.m.18k Da and E.m.16 k Da for immunodiagnosis of alveolar echinococcosis, and E.m.18k Da in particular.
