摘要
为研究ApoE基因对中国人非胰岛素依赖型糖尿病(NIDDM)患者血脂水平的影响,应用聚合酶链反应(PCR)扩增出ApoE基因第4外显子内的292bp片段,继以限制性内切酶HhaⅠ酶解消化,最后行8%聚丙烯酰胺凝胶电泳,根据电泳带型判断ApoE基因型。共检测了112例非胰岛素依赖型糖尿病患者的ApoE基因型。通过比较不同ApoE基因型与血脂水平的关系发现:ApoE基因多态性与TG,HDL-C,ApoAⅠ,ApoB及Lp(a)水平无相关性,而与TC(P=0.0029)及LDL-C(P=0.0021)水平相关,携带ε4等位基因者具有较高的TC及LDL-C水平,而携带ε2等位基因者具有较低的TC及LDL-C水平。
To study the effects of Apo E gene on the serum lipid levels in Chinese with non insulin dependent diabetes mellitus(NIDDM), a 292 base pair DNA fragment in the fourth exon of ApoE gene was amplified by polymerase chain reaction (PCR), analyzed by digestion with restriction enzyme Hha Ⅰ, and followed by 8% polyacrylamide gel electrophoresis of the cleavage products, subsequently the Apo E genotype was determined in 112 NIDDM patients. Comparing the values of the serum lipids among NIDDM patients with different genotypes, we found no impact of the various Apo E genotypes on the serum levels of TG, HDL C, ApoA I, Apo B and Lp(a), but Apo E gene polymorphism was significantly associated with TC( P =0.0029) and LDL C( P =0.0021) in NIDDM patients. NIDDM individuals carrying ε4 allele were associated with increased concentrations of TC and LDL C, while those carrying ε2 allele were associated with decreased concentrations of TC and LDL C.
