摘要
目的:观察蝙蝠葛碱(Dau)对培养的平滑肌细胞经内皮素诱导增殖的影响,探讨其抗动脉粥样硬化作用的机制。方法:以细胞计数及氚标胸腺嘧啶核苷参入试验观察细胞增殖及脱氧核糖核酸(DNA)合成;以流式细胞技术观察细胞周期。结果:内皮素明显促进平滑肌细胞增殖及脱氧核糖核酸合成;Dau对此有显著抑制作用。结论:Dau可抑制内皮素诱导的平滑肌细胞增殖,这可能与其抗动脉粥样硬化作用有关。
Objective:Smooth muscle cells (SMCs) proliferation is a key event in atherogenesis,with which endothelin (ET) is associated.To explore the antiatherosclerotic mechanism of dauricine(Dau) by examining its effect on cultured arterial SMCs proliferation induced by ET in vitro. Methods:Cell counting and 3H thymidine incorporation were used to observe SMCs proliferation and DNA synthesis.Cell cycle of SMCs was observed by flow cytometric analysis(FCA). Results:ET(0.1 μmol/L) markedly stimulated SMCs proliferation and DNA synthesis( p <0.01),which could be inhibited by Dau(10~100 μmol/L) significantly( p< 0.01). Conclusion:Dau has an inhibitory effect on SMCs proliferation induced by ET, which might implicate in antiatherosclerotic effect.
