视网膜缺氧及复氧中Caspase-1的活性变化

摘要 目的研究Caspase-1是否参与视网膜缺氧及复氧后视网膜神经元的损伤,为视网膜缺血性损伤的治疗提供实验资料。方法21只成年小鼠用升高眼内压法致视网膜缺血缺氧,于再灌注复氧后1 h、3 h、6 h、1 d、3 d、7 d处死动物。冰冻切片进行Caspase-1免疫组化及TUNEL染色。结果正常视网膜没有Caspase-1阳性细胞。复氧后1 h,视网膜节细胞层及内核层有许多阳性细胞;3 h后Caspase-1阳性细胞继续增加,于6 h达高峰,1 d时阳性细胞数减少。TUNEL阳性细胞开始出现于复氧后3 h,1 d时达高峰。Caspase-1免疫组化与TUNEL双重标记显示缺血后6 h及24 h少量散在阳性细胞。结论在视网膜缺氧及复氧损伤时节细胞层及内核层Caspase-1阳性细胞明显增加,且时相提前于TUNEL阳性细胞,提示Caspase-1早期激活可能具有启动视网膜神经元凋亡过程,通过抑制其活性可保护视网膜神经元免受损伤。
