1-(2,6-二甲基苯氧基)-2-(3,4-二甲基苯乙氨基)丙烷盐酸盐对心肌肥厚大鼠左室肌酸激酶及同工酶谱的影响

摘要 研究1-(2,6-二甲基苯氧基)-2-(3,4-二甲基苯乙氨基)丙烷盐酸盐(DDPH)对心肌肥厚的逆转作用和对左室肌酸激酶(CK)及同工酶谱的影响。方法用部分狭窄腹主动脉的方法造成大鼠心肌肥厚模型,术后4w开始igDDPH25和50mg/kg·d-1,连续8w。结果术后12w,各组大鼠体重无显著性差异,但模型组左室重/全心重,全心重/体重分别比对照组高39.0%和36.9%,左室CK活性各组无明显差异,但模型组MB同工酶比对照组高3.4倍,MM同工酶降低143.6%.而2个给药组则明显使左室重/全心重和全心重/体重逆转,并减少肥厚心肌MB同工酶相对百分含量。结论DDPH可逆转部分狭窄腹主动脉所致心肌肥厚,同时可逆转心肌肥厚时肌酸激酶同工酶谱的变化。 Aim To investigate the reversing effects of 1 (2,6 di methyphenoxy) 2 (3,4 dimethyoxyphenylethylamino)propane hydrochloride(DDPH) in CPK activity and CPK MB isoenzymes of ventricular hypertrophic rats.\ Methods\ The model of rat cardiac hypertrophy was made by partly narrowing abdominal aorta.\ The rats were divided into four groups at random:(1)control group; (2) model group;(3)low dosage of 1 (2,6 di methyphenoxy) 2 (3,4 dimethyoxyphenylethylamino)propane hydrochloride(DDPH)(25 mg·kg -1 ·d -1 ) group;(4)higher dosage of DDPH(50 mg·kg 1 ·d -1 ) group.\ Results\ Four weeks after operation, the rats were given drugs four 8 weeks.\ 12 weeks later, it was founds that left ventricle weight(LVW)/whole heart weight(WHW) and WHW/body weight(BW) were increased by 39.0% and 36.9% than that in control group.\ There was no significant difference in CPK activity in left ventricle among four groups, but the isozyme type changed.\ MB isozyme of CPK in model group was 4.4 times higher than in control group, but MM isozyme decreased 143.6%.\ Subunit B increased 2.8 times, subunit M decreased 40%.\ Conclusion\ that DDPH can reverse cardiac hypertrophy of rats induced by partly narrowing abdominal aorta, but also reverse the changes of CPK activity and its isozymes.
