单纯疱疹病毒介导GALV致融性糖蛋白对人肺腺癌裸鼠移植瘤的抑制作用

摘要 目的探讨Ⅰ型单纯疱疹病毒(HSV-Ⅰ)载体介导的长臂猿白血病病毒致融性外膜糖蛋白(GALV.fus)基因转导肺腺癌细胞的体内抗肿瘤作用及建立人肺腺癌皮下瘤裸鼠模型的最佳方案。方法将重组HSV-Ⅰ载体质粒通过脂质体导入非洲绿猴肾细胞(Vero)中包装制备重组HSV-Ⅰ病毒,通过细胞悬液接种或组织块移植法构建人肺腺癌细胞(A549)裸鼠皮下移植瘤模型,将重组HSV-Ⅰ病毒注入裸鼠肺癌瘤体中观察其体内抗肿瘤作用。结果成功包装重组HSV-Ⅰ病毒,病毒对肺癌细胞有明显的杀灭作用且对裸鼠皮下肺癌转移灶抑制明显。结论细胞悬液接种和套管针组织块接种均为建立裸鼠荷瘤模型理想方法;GALV.fus基因对肺癌细胞有强效的抗肿瘤作用,明显抑制人肺腺癌裸鼠移植瘤的生长,为临床肺癌治疗探索出一种新型基因治疗方法。
