西洋参叶二醇组皂苷对血管平滑肌细胞增殖及凋亡的影响

Effect of PQDS on proliferation and apoptosis-gene expression of VSMC

目的观察西洋参叶二醇组皂苷(PQDS)对血管紧张素Ⅱ(AngⅡ)诱导的血管平滑肌细胞(VSMC)增殖及凋亡的影响。方法将组织贴块法培养的大鼠胸主动脉VSMC随机分为5组:空白组、模型组及PQDS 25,50,100mg/L组。应用MTT法检测各组VSMC的生长率,以流式细胞术检测细胞周期构成比和增殖指数(PI),以TUNEL法检测细胞凋亡率。结果PQDS可明显减低VSMC的增殖能力,使VSMC的G_0/G_1期构成比显著升高,PI值显著下降;亦能明显增加VSMC的凋亡率。结论PQDS具有抑制VSMC增殖及诱导其凋亡作用。

Objective To investigate the effect of Panax Quinquefolium Diolsaponins （PQDS） on proliferation of vascular smooth muscle cell （VSMCs） and the apoptosis induced in angiotensin Ⅱ （ Ang Ⅱ ）. Methods VSMCs cultured by tissue-sticking method were randomly divided into control, model and PQDS 25, 50 and 100 mg/L groups. The cell proliferation ability was measured by MTT assay, the cell generation cycle and prolifation index were measured by Flow cytometery, apoptosis rate of VSMC was measured by TUNEL method. Resuits PQDS inhibited significantly the proliferation of VSMC, up-regulated significantly the constituent ratio of VSMC G0/G1 cell cycle and down-regulated the proliferation index. PQDS also increased significantly apoptosis rate of VSMC. Conclusions PQDS could inhibit significantly the proliferation of VSMC, and induce the apoptosis of VSMC.
