MMP-9和TGF-β_1在STZ-糖尿病大鼠晶状体上皮细胞的表达 被引量:3

Expression of transforming growth factor β_1 and matrix metalloproteinase 9 in lens epithelial cells of streptozotocin-diabetic rats

摘要 目的探讨基质金属蛋白酶-9(matrix metalloproteinase9,MMP-9)和转化生长因子-β1(transforming growth factor β1,TGF-β1)在链脲佐菌素(streptozotocin,STZ)诱导的糖尿病大鼠晶状体上皮细胞中的表达及意义,了解MMP-9与TGF-β1在糖尿病性白内障发病中的作用。方法8周龄SD大鼠60只随机分为对照组和实验组,给予实验组大鼠20g.L-1STZ溶液55mg.kg-1一次性腹腔注射,诱发糖尿病性白内障。每周裂隙灯观察大鼠晶状体变化。采用免疫组织化学方法检测2周末、4周末、8周末时MMP-9及TGF-β1在大鼠晶状体上皮细胞中的表达。结果对照组大鼠晶状体始终保持透明,实验组大鼠晶状体逐渐混浊。实验组MMP-9和TGF-β1的阳性表达高于对照组,差异有统计学意义(P<0.05);实验组MMP-9和TGF-β1阳性表达逐渐增高,各时段间差异有统计学意义(P<0.05)。MMP-9和TGF-β1的阳性表达呈正相关(r=0.937,P<0.01)。结论MMP-9和TGF-β1的表达增高可能在糖尿病性白内障的发生、发展中有重要作用。  Objective To investigate the expression and significance of transforming growth factor β1(TGF-β1) and matrix metalloproteinase 9 (MMP-9) in lens epithelial cells(LECs) of streptozotocin-diabetic rats,and study the effects of TGF-β1 and MMP-9 on rats with diabetic cataract.Methods Sixty SD rats which were 8-week-old were randomly divided into experimental and control groups.Rats of experimental group were induced diabetic cataract for intraperitoneal injection of 20 g·L^-1streptozotocin (55 mg·kg^-1in 0.1 mol·L^-1sodium citrate buffer).Lens were examined using a slit lamp once a week.Immunohistochemical streptavidin-peroxidases method was used to detect the expression of MMP-9 and TGF-β1 in experimental and control lens at the end of 2 weeks,4 weeks and 8 weeks.Results The lens of control group were always completely transparent throughout the period of experiment,and the lens of experimental group became opaque gradually.The expression of MMP-9,TGF-β1 of experimental LECs were significantly higher than that of control LECs(P〈0.05);During the progress of cataract,the expression of MMP-9,TGF-β1 gradually increased(P〈0.05).The expression of MMP-9 was positively correlated with that of TGF-β1(r=0.937,P〈0.01).Conclusion The expression of MMP-9,TGF-β1 may play a critical role during the development of diabetic cataract.
