摘要
目的探讨衰老大鼠阴茎组织转化生长因子β(TGF-β)基因表达与补肾通络疏肝方在阴茎勃起中的关系。方法将32只雄性大鼠随机分为衰老组、补肾通络疏肝方组、丙酸睾酮组与对照组(2月龄大鼠),其中补肾通络疏肝方组和丙酸睾酮组分别给予补肾通络疏肝方及丙酸睾酮治疗,于24 d取阴茎组织,用PCR检测其TGF-βmRNA的表达。结果衰老组TGF-βmRNA的表达较其他3组增强,而表现TGF-βmRNA减弱的组,由少到多依次排定为对照组、丙酸睾酮组及补肾通络疏肝方组;对照组、丙酸睾酮组及补肾通络疏肝方组其TGF-βmRNA内控量较少,与衰老组比较有显著性差异(P<0.01),丙酸睾酮组及补肾通络疏肝方组之间无显著性差异(P>0.05)。结论补肾通络疏肝方可能通过调节TGF-βmRNA表达而在阴茎勃起中发挥重要作用。
Objective To investigate the relationship between the expression of transforming growth factor-β (TGF-β) mRNA and Bushentongluoshugan Formula in penile erection in senescent rats. Method All 32 mature male rats were randomly divided into: the senescent rat group (group 1 ), Bushentongluoshugan Formula group ( group 2 ), testosterone propionate group ( group 3 ) and control group (group 4, the rats were 2 months old). On the 24th day the penile tissue was taken from the rats and the expression of TGF - β mRNA was detected by PCR technique. Result In group 1 the expression of TGF-β mRNA was increased than those of other three groups. The groups with decreased the expression of TGF-β mRNA were ordered as follow ascent sequence: group4, group 1, group 3, group 2. The controlled amount of TGF-β mRNA in group 4, 3 and 2 was lower and had a significant difference respectively compared with that of group 1 (P 〈 0.01 ). There was no significant difference between group 3 and 2 ( P 〉 0.05 ). Conclusion Bushentongluoshugan Formula may play an important role in the penile erection through regulating the expression of TGF-β mRNA.
