急性胰腺炎大鼠胰腺细胞膜流动性、钙泵活性的变化及四甲基吡嗪的影响

Alteration of pancreatic cell membrane fluidity and calcium pump in rats' experimental acute pancreatitis and effect of tetramethylpyrazine treatment

目的：观察大鼠急性胰腺炎（ＡＰ）时胰腺细胞膜流动性、钙泵活性的变化及四甲基吡嗪对其的影响。方法：动态测定大鼠急性胰腺炎用四甲基吡嗪治疗前后胰腺细胞膜荧光偏振度、钙泵活性、丙二醛含量、淀粉酶崐活性，并测定胰组织钙离子含量。结果：四甲基吡嗪治疗组，钙泵活性较非治疗组增高，荧光偏振度、丙二醛含量、淀粉酶活性和钙离子含量则降低。在ＡＰ非治疗组，相关分析显示荧光偏振度的增高与膜悬液中丙二醛含量，钙离子含量则成负相关，Ｐ＜０．０５．结论：用四甲基吡嗪能提高ＡＰ大鼠胰腺细胞的钙泵活性，降低膜荧光偏振崐度，减轻细胞内钙超载，从而提高了细胞膜的流动性，减轻过氧化反应。

Objective: To study the alteration of pancreatic cell membrane fluidity and calcium pump in rats' experimental acute pancreatitis with special reference to change after tetra- methylpyrazine treatment. Methods: Pancreatic cell membrane Ca -Mg -ATPase ac- tivity,fluorescence polarization, MDA content, Ca2. and amylase activity were studied. Results: Pancreatic cell membrane fluorescence polarization, MDA content, Ca2+ and amylase activity were increased, while Ca2+-Mg2+ -ATPase activity was decreased after induction of AP. By linear correlation analysis, it was found that there were strong negative correlation between Ca2+-Mg2+ -ATPase activity and MDA content, Ca2+ or amylase activity, and positive correlation between fluorescence polarization and MDA content, Ca2+ or amylase activity, In treated group with TMP, pancreatic cell membrane fluorescence polarization, value of MDA, Ca2+ and amylase activity were much lower, Ca2+-Mg2+-ATPase activity was much increased than that in untreated group(P< 0.05). Conclusion: Decreased pancreatic cell membrane Ca2+-Mg2-ATPase activity and fluidity in rats with AP might be caused by lipid peroxidation reaction and intracellular Ca2+ overload. TMP showed its beneficial effect on the preservation of pancreatic cell membrane, which was related to suppression of lipid peroxidation reaction and intracellular Ca2+ influx.
