不同产地丹参药材的ISSR分析与鉴别 被引量:24

摘要 目的对不同产地丹参药材的亲缘关系进行分析研究,建立具有鉴别意义的DNA指纹图谱。方法采用ISSR分子标记对丹参药材的基因组DNA进行PCR扩增,利用NTSYS-pc2.1和POPGENE 1.32软件分析不同产地药材间的亲缘关系,构建树系图。结果从66条ISSR引物中筛选出两条具有鉴别意义的多态性引物,在11个不同产地的丹参药材中共得到32位点,1条是单态的,31条是多态的,多态性位点达96.88%,遗传距离在0.1699~1.0678,平均为0.4712。结论不同产地的丹参药材间遗传分化明显,具有丰富的遗传多样性,但是遗传分化与地理关系没有表现出相关性。ISSR标记可以作为不同产地丹参药材鉴别的有效分子标记。 Objectve To study the genetic relationships of Radix et Rhizoma Salvia miltiorrhizae from different habitats, and to establish the DNA fingerprints for identification. Methods PCR was performed with the genomic DNA of Radix et Rhizoma Salvia miltiorrhizae by ISSR marker, the genetic relationships were analyzed by software POPGENE 1.32 and the DNA molecular dendrogram was set up by the software NTSYS - pc 2.1. Results The two effective primers for identification were chosen from 66 ISSR primers. A total of 32 loci were amplified from Radix et Rhizoma Salvia miltiorrhizae from 11 habitats by two effective primers. In the total 32 amplified bands, 31 (96, 88 % ) were polymorphic. The genetic distances were from 0. 169 9 to 1. 067 8, with an average of 0. 471 2. Conclusion There is abundant genetic diversity among Radix et Rhizoma Salvia miltiorrhizae and the genetic relationships do not show coherent with geographical distance. The ISSR markers can be used for the identification of Radix et Rhizoma Salvia miltiorrhizae from different habitats.
