转基因抗虫棉晋棉44号选育研究及高产栽培要点 被引量:2

摘要 采用农杆菌介导转化法,将CryL4c3导入受体棉花品系冀合492中,获得再生植株。经PCR扩增分析,得到和预期长度一样大小的743pb目的基因片段,初步证明外源基因已整合到棉花基因组中。进行PCR-Southern杂交检测结果,所有PCR阳性植株产物电泳带均显示出较强信号,进一步说明外源Bt基因已整合到棉花体内。生物检测结果,转基因棉花对棉铃虫具有很高抗性,苗期、蕾期、开花期高抗株率分别达80%-96%,说明转化CryLAc3可以使受体棉花具有很高的杀虫活性。通过连续选择和自交纯合及南繁加代,得到综合性状优良的品系2001pb-3。参加山西省中熟棉花区域试验,2005年通过审定,定名为晋棉44号。晋棉44号农艺性状优良,高产优质。 Foreign genes CrylAc3 were introduced into conventional cotton cultivars Jihe492 by Agrobacterium tumefaciens - mediated transformation. Regenerated plants have been obtained. Foreign target - gene about 743 bp was detected through PCR molecular detection and the results indicated that the foreign genes had been transferred into cotton plants. All electrophoretic strips of PCR electropositive plants have a strong signal in PCR - Southern detection. The results of biologic - detection indicated that the transformed cotton has a higher insect - resistance, respective to 80% -96% in seedling period, bud period and anthesis. The better lines 2001pb -3 has been obtained by series select, inbred and south - increase. It attended the medium -ripe cotton region test in Shanxi province, which was authorized in 2005 and named Jinmian 44. Jinmian 44 is better in agriculture character and yield.
