摘要
目的:观察尼古丁及卡巴胆碱对谷氨酸诱导的小胶质细胞增殖的影响.方法:培养的小胶质细胞系,加入不同浓度的尼古丁、卡巴胆碱及谷氨酸,用MTT方法检测细胞活性,观察对小胶质细胞数量的变化.结果:培养的小胶质细胞提示,尼古丁及卡巴胆碱显著减少谷氨酸诱导小胶质细胞数量的增多,有剂量依赖效应,10μmol/L浓度效果最为显著.结论:尼古丁及卡巴胆碱可显著抑制谷氨酸诱导小胶质细胞活化的增殖.
AIM: To observe the effects of nicotine and carbachol on the proliferation of microglia induced by glutamate. METHODS: The number of cultured microglia treated with different doses of nicotine, carbachol and glutamate respectively was determined by MTT assay. RESULTS: Nicotine and carbachol decreased the number of microglia in the presence of glutamate in a dose-dependent manner, and most significantly at 10 μmol/L concentration of nicotine or carbachol. CONCLUSION : Nicotine and carbachol inhibit significantly microglial proliferation induced by glutamate.
